Methodology
Direct Microscopic Examination of Stained Smear
Useful for determining the presence or absence of bacteria,
yeast, WBCs, and epithelial cells.
Note:
Gram stains are automatically included in the following routine cultures:
- body fluid
- bronchial washings
- sputum
- wound
The following cultures may be ordered with or without Gram stains:
- abscess
- aerobic
- ear
- eye
- genital
- nares
- nasopharyngeal
- throat
If a culture is also desired, order specific culture with Gram stain combination. Be sure to submit enough material for both Gram stain and culture procedures.
Specimen Requirements
Gram stain may be performed on any site or source suitable for culture. See individual culture listings for appropriate specimen collection instructions.

Reference Values
Dependent upon site/source of specimen.
Critical value (automatic call-back): bacterium present on
cerebrospinal fluid or other sterile body fluid
Note:
- Gram stain is not as sensitive or specific as culture.
- If organism identification and susceptibilities are desired, a culture must be requested.
- False-negative results occur with Gram stains due to low number of organisms present in the specimen.
