Methodology
Direct Microscopic Examination
Useful for examination for the presence of motile protozoa such as trichomonas and for the detection of yeast. The wet mount is negative in up to 30% of women with trichomonas infections.

Collection Instructions:
- Patient should not have urinated for at least 1 hour prior to specimen collection.
- Insert small-tipped specimen swab 2 cm to 4 cm into urethra, rotate, and leave in place for 2 seconds to ensure adequate sampling.
- Withdraw swab.
- Place swab in sterile container with 0.5 mL of sterile saline.
- Mix swab well with saline and discard swab.
- Label container with patient’s legal name, date and actual time of collection, and type of specimen.

Collection Instructions:
- Remove excess mucus from exocervix and discard.
- Insert a large culture swab into vagina and swab cervix, endocervix, or vaginal wall.
- Place swab in sterile container with 0.5 mL of sterile saline.
- Mix swab well with saline and discard swab.
- Label container with patient’s legal name, date and actual time of collection, and type of specimen.
