Methodology

Real-Time Polymerase Chain Reaction (PCR)/DNA Probe Hybridization

(PCR is utilized pursuant to a license agreement with Roche Molecular Systems, Inc.)

 

Useful for rapid (qualitative) detection of Bordetella pertussis and Bordetella parapertussis DNA.

Specimen Requirements

Container/Tube:

 

Not Acceptable: Nares swab, Throat swab, calcium alginate, cotton-tipped swab or swabs with wood shafts.

 

PreferredBD Eswab collection kit includes the transport device with Nasopharyngeal swab, #65504

Acceptable -  Nasopharyngeal Swab in Universal Viral Transport Media

Tube/Container: UTM Collection Kit, (Supply order - #0075681)

 

Specimen Volume: Entire specimen

 

Collection Instructions:

  1. Estimate distance to nasopharynx: prior to insertion, measure distance from corner of nose to front of ear and insert swab ONLY half this length.
  2. Tilt patient’s head back slightly to straighten passage from front of nose to nasopharynx, to make insertion of swab easier.
  3. Insert swab into 1 nostril.
  4. Gently insert swab along medial part of septum, along base of nose, until it reaches posterior nares. Gentle rotation of swab may be helpful. (If resistance is encountered on 1 side, try other nostril, as patient may have a divided septum.)
  5. Insert swab into the green top transport device (to preserve specimen), snap off swab at the score mark and reattach screw on top.
  6. Label container with patient’s legal name, date and actual time of collection, and type of specimen.
