Methodology
Semiquantitative Tube Titer
When a clinically significant antibody has been identified in a pregnant woman, this test is used to determine antibody concentration and to monitor changes in antibody levels throughout the pregnancy.
Specimen Requirements
Submit only 1 of the following specimens:
Preferred:
Pink-Top Tube
Container/Tube: 2 (6 mL) Pink-top (EDTA) “crossmatch” tube(s)-Plain, red-top tube or serum gel tube is not acceptable.
Specimen Volume: 12 mL of whole blood
Collection Instructions: Forward promptly in original tube(s).
Note: Include report of diagnosis and history of transfusions, pregnancy, and drug therapy.
Alternate:
Lavender-Top Tube
Container/Tube: Lavender-top (EDTA) tube(s)-Plain, red-top tube or serum gel tube is not acceptable.
Specimen Volume: 12 mL of whole blood
Collection Instructions: Forward promptly in original tube(s).

Reference Values
Results are reported as the reciprocal of the highest dilution of serum at which agglutination is observed.
A titer ≥16 is considered significantly elevated.
